Low Melting Point Agarose (CAS 39346-81-1, Gelling 26 to 30 C, Melting below 65 C)

Cat No:MCH-0033

Description

Modified low melting point agarose with improved molecular sieving and band clarity, melting at about 65.5 C so recovered nucleic acid bands can be remelted below nucleic acid melting points.

Storage Conditions Store sealed at room temperature; five-year validity
Intended Use Separation and recovery of nucleic acid fragments larger than 1000 bp, cloning directly from remelted gel slices
Target Industries Molecular biology research, cloning and probe preparation workflows
Recommended Applications DNA fragment recovery, radiolabelled probe preparation, plaque lifts and radial immunodiffusion protein assays
Recommended Use Level Cast gels at 1 to 2 percent for fragments above 1000 bp
Appearance of Solution Melt fully by boiling or microwave; remelt recovered gel slices at 65 C for downstream use
Handling & Safety Handle as a fine powdered material; avoid dust formation and wear gloves, mask and eye protection
Chemical Class Purified agarose galactan hydrocolloid
Structural Unit Linear chain of alternating 1,3-linked beta-D-galactose and 1,4-linked 3,6-anhydro-alpha-L-galactose
Functional Groups Hydroxyl groups and bridged 3,6-anhydro rings forming helical gel junctions
Ionic Character Neutral polysaccharide with minor anionic sulfate impurities
Key Features Melting below the denaturation temperature of most nucleic acids enables direct enzymatic use of recovered DNA; Gelling point 26 to 30 C at 1.5 percent; melting point 65 C or below at 1.5 percent
Functional Benefits Shortens cloning workflows by allowing ligation-ready DNA straight from remelted slices
Typical End Products Agarose gels for DNA electrophoresis, blotting and fragment recovery
Formulation Compatibility Compatible with restriction digestion and ligation after equilibration in suitable buffer
Preparation & Dissolution Guidance Hydroxyethylation modification of purified agarose
Reactivity & Derivatization Helix-coil transition on heating and cooling; sulfate hydrolysis only under strong acid
Stability Notes Five years sealed at room temperature
Usage Precautions Molten gel can bump violently; wear heat protection and lab coat
Environmental Profile Biodegradable polysaccharide
Analytical Methods Gel strength, gelling and melting points, EEO and sulfate per product specification
