Carbohydrate Esterases

An acetyl group carries just over forty-three daltons and occupies two or three atoms of space on a sugar ring. Nothing about it looks important. Scatter a few hundred along a polymer and they decide whether the material gels, whether a hydrolase can reach the backbone at all, and whether the fermentation downstream survives the acetic acid that comes off when they are stripped. Substituents this small are where polysaccharide processes quietly go wrong, and they are also the cheapest thing to fix once you have the right catalyst.

Eata Marisource supplies carbohydrate esterases that change those substituents without shortening the chain: acetyl xylan esterases spanning the CE1 to CE7 families, metal-dependent CE4 deacetylases active on chitin and chitooligosaccharides, pectin and rhamnogalacturonan acetylesterases, glucuronoyl esterases for lignin-carbohydrate linkages, and marine-derived esterases that act on bacterial alginate. Production is documented host by host, purification is taken to a declared level, and the release test measures activity on a named substrate rather than on a borrowed chromogenic one.

Where the Acetyl Groups Sit

— 01 — and what each one is doing to your material

The same substituent behaves differently depending on the polymer it hangs from, so the first conversation is always about the substrate and never about the enzyme catalogue.

Polymer Position of the substituent Effect on behaviour What removal buys you
Xylan and related hemicelluloses O-2 and chiefly O-3 of backbone xylosyl residues Steric shielding of the backbone, so endo-xylanases stall; hydrolysis then releases acetate Hydrolases regain access and the acetate load in the next vessel falls at the same time
Bacterial alginate O-2 and/or O-3 of mannuronate residues Epimerases cannot convert acetylated residues, and neighbours are shielded too; most lyases are also blocked C-5 epimerases can then build G-blocks, raising gel strength at unchanged chain length
Chitin and chitooligosaccharides N-acetyl on the glucosamine nitrogen Crystallinity, insolubility and poor reactivity A soluble, amine-bearing material whose degree of deacetylation can be held where you want it
Pectin and rhamnogalacturonan O-acetyl on galacturonic acid, alongside methyl esters Blocks polygalacturonase attack and shifts setting behaviour Cleaner enzymatic depolymerisation and adjustable setting performance
Glucuronoxylan bound to lignin Ester link between 4-O-methyl-glucuronic acid and lignin Locks the carbohydrate into the lignin network Carbohydrate released from lignin-carbohydrate complexes instead of being lost with the residue

One clarification saves a good deal of wasted money. Brown-algal alginate reaches the market essentially unacetylated; the acetylated polymer is the bacterial material made by Azotobacter and Pseudomonas species, plus the alginate inside biofilms. If your feedstock is seaweed-derived, tell us before ordering a deacetylase, because the right answer is more likely a lyase or an epimerase, and we would rather say so at the start.

Reading the Family Map

— 02 — why CE4 covers both a xylan esterase and a chitin deacetylase

Families are an index, not a specification. Two members of CE4 can differ from each other far more than either differs from an enzyme in another family, and the binding modules appended to the catalytic domain often decide whether an esterase works on your material at all. The table is the short version; the detail that matters is in the catalytic machinery column, because it tells you which reagents will silence the enzyme.

Family Activities encountered Representative EC Catalytic machinery Buying note
CE1 acetyl xylan esterase, feruloyl and cinnamoyl esterase EC 3.1.1.72, EC 3.1.1.73 serine-histidine-aspartate triad, alpha/beta hydrolase fold Usual first choice when bound phenolic acids are the target
CE2 acetyl xylan esterase, acetyl beta-mannan esterase EC 3.1.1.72, EC 3.1.1.6 SGNH fold plus a separate jelly-roll domain Broader tolerance, often gentler on acetylated mannan
CE3 acetyl xylan esterase EC 3.1.1.72 serine triad Narrower use, mostly biomass programmes
CE4 acetyl xylan esterase, chitin deacetylase, chitooligosaccharide deacetylase, peptidoglycan deacetylase EC 3.1.1.72, EC 3.5.1.41 metal-dependent NodB domain, usually zinc, cobalt in some members Chelators stop it dead; see the buffer section before you formulate
CE5 acetyl xylan esterase, cutinase EC 3.1.1.72, EC 3.1.1.74 serine triad Cutinase activity is useful on cutin-containing feedstock and unwanted elsewhere
CE6 acetyl xylan esterase EC 3.1.1.72 serine-histidine-glutamate Small and robust, a good first screening candidate
CE7 acetyl xylan esterase EC 3.1.1.72 serine triad, commonly hexameric Stable, but often a narrower substrate range
CE8 pectin methylesterase EC 3.1.1.11 right-handed beta-helix Changes setting behaviour rather than removing acetyl groups
CE9 N-acetylglucosamine 6-phosphate deacetylase EC 3.5.1.25 metal-dependent distorted barrel For aminosugar catabolism studies rather than polymer work
CE11 UDP-3-O-acyl-N-acetylglucosamine deacetylase EC 3.5.1.- zinc-dependent two-domain sandwich Research tool in lipid A biosynthesis work
CE12 pectin acetylesterase, rhamnogalacturonan acetylesterase EC 3.1.1.86 serine triad The choice when the substrate is pectic rather than a xylan
CE13 pectin acetylesterase — not yet resolved in detail Limited supply; screened on request
CE14 deacetylases acting on N-acetylated aminosugar derivatives EC 3.5.1.89 alpha/beta hydrolase fold with an N-terminal Rossmann motif Overlaps CE4 in practice, so choose by measured specificity
CE15 glucuronoyl esterase — serine-type Releases carbohydrate from lignin-carbohydrate complexes

Further families have been added to the classification since, but they are sparsely characterised and rarely worth quoting in a purchase specification; where one is relevant to your substrate we will say so and explain what has actually been measured.

Two Cases That Come back Repeatedly

— 03 — bacterial alginate, and chitin on the way to chitosan

Bacterial alginate: stronger gel, same chain length

Every route to a higher G-content carries a cost. A lyase shortens the chain, so viscosity drops with it. Hot alkali removes acetyl but also depolymerises and darkens the product. Deacetylating first and epimerising second is the only sequence that lifts G-content while leaving molecular weight alone, and the reason is structural: epimerases cannot act on an acetylated residue, and residues neighbouring an acetyl group are shielded as well. In practice the deacetylation step is what unlocks the epimerase rather than a standalone improvement.

Availability is narrower than most buyers expect. The alginate deacetylase activity described in the literature comes from a Pseudomonas syringae enzyme whose separate N-terminal module belongs to the SGNH hydrolase superfamily, and it is that module which lets the same protein work on acetylated substrate before epimerising it. Treating bulk bacterial alginate at process scale is therefore usually a development project rather than something bought off a shelf, and we would rather frame it that way at the start.

Chitin to chitosan: the pattern matters as much as the percentage

Caustic gives you one number - eighty-five per cent deacetylated, say - and a random distribution of the remaining acetyl groups. Enzymatic deacetylation gives you a second lever, because characterised CE4 members are known to follow processive, multiple-attack or patterned mechanisms depending on the enzyme. Two chitosans sharing an average degree of deacetylation but carrying different patterns differ in solubility, viscosity, metal binding and behaviour against other polyanions, so the average alone does not describe the product.

The constraint worth knowing before you budget: crystalline chitin is a poor substrate. Activity improves sharply once the material has been mechanically opened, swollen, or converted to soluble chitooligosaccharides, and which of those applies decides whether an enzymatic route is realistic for your stream. That assessment is part of the enquiry, not an afterthought.

What the Numbers on a Certificate Mean

— 04 — chromogenic units, natural substrates and the chelator trap

Esterases are sold with more borrowed assays than any other enzyme class. p-Nitrophenyl acetate is cheap and quick, and it measures something your polymer has never heard of: lots that look identical against that substrate can differ several-fold once they meet acetylated xylan. We report the chromogenic figure because buyers ask for it, and we also report activity on a named natural substrate with the released acetate quantified, because only the second number predicts a reactor.

  • Unit definition: one unit releases one micromole of acetate per minute from the named substrate at the stated pH, temperature and ionic strength. The p-nitrophenyl acetate figure appears separately and is never presented as a release rate on polymer.
  • Cofactor: CE4 members hold a metal cofactor, usually zinc and in some characterised members cobalt, coordinated through an aspartate-histidine-histidine triad. EDTA, citrate or a phosphate-heavy buffer will strip it. Serine-type esterases instead answer to serine hydrolase inhibitors, so treating an unknown preparation with the wrong inhibitor is a fast way to misclassify what you bought.
  • Thermal behaviour: thermostable members are held for process work in the 60 to 70 degrees Celsius range, while cold-adapted marine members suit streams that cannot be heated. A half-life curve at your operating temperature is included on request rather than a single optimum-temperature point.
  • Side activities: each lot is screened for protease and for glycosidase contamination, since a trace hydrolase quietly changes chain length while you are measuring acetate release and attributing the change to something else.
  • Excipient: lots are filled with trehalose or mannitol rather than bulking protein, so reconstitution is quick and the declared weight is closer to active material.

Acetate itself is easy to follow and easy to lose. Released acetate can be quantified enzymatically, by ion chromatography, or by high performance liquid chromatography after the protein has been removed and the sample concentrated, and the answer only means something if the sampling quench is fast enough that no further enzyme action occurs in the tube. We supply the quench and cleanup protocol with the certificate rather than leaving it to be rediscovered.

Where the Economics Actually Move

— 05 — seven places an esterase pays for itself

  • Biomass conversion: released acetate is a fermentation inhibitor and acetyl groups block hydrolase access, so one deacetylation step raises sugar release and lowers inhibitor load in the same unit operation instead of trading one against the other.
  • Alginate upgrading: deacetylation ahead of epimerisation lifts G-content while molecular weight is untouched, which is the only combination available when gel strength and viscosity both have to hold.
  • Chitosan production: the enzymatic route avoids the darkening and chain scission of hot caustic, and gives a deacetylation pattern rather than only an average, so the product specification can say more than one number.
  • Pulp and fibre processing: esterases are useful accessory activities wherever acetyl groups limit chemical access to fibre; the benefit shows up as milder downstream conditions rather than as a headline yield figure.
  • Bound phenolics: feruloyl and cinnamoyl esterases liberate bound phenolic acids from bran, pulp and pressed residues, which turns a waste fraction into a recovered ingredient stream.
  • Analytical work: measuring acetate released by a defined enzyme is the cleanest way to determine degree of acetylation, and partial enzymatic deacetylation followed by liquid chromatography tandem mass spectrometry lets the substituent pattern be mapped rather than averaged.
  • Biofilm and cell wall research: partial deacetylation of poly-N-acetylglucosamine in biofilm matrices is a recognised research tool, and deacetylases are also used to remodel peptidoglycan in cell wall studies.

Formats and Handling

— 06 — presentation, pack sizes and the buffer mistakes to avoid

Presentation Best for Pack sizes Storage Buffer caution
Lyophilised powder Reproducible bench work and long-term storage 10 mg to 100 g −20 °C, sealed with desiccant Metal-dependent members: avoid EDTA and citrate in the reconstitution buffer
Stabilised liquid Feasibility trials and immediate dosing 1 mL to 50 mL 2 to 8 °C short term, −20 °C in glycerol for longer Check the stated glycerol content before adding it to a cold process stream
Immobilised preparation Repeated batch use and packed-bed contactors 10 to 500 mL settled bed 2 to 8 °C, never frozen Coupling chemistry is chosen around the catalytic metal where one is present
Process-grade bulk Plant trials and routine production kilogram lots, matched activity Chilled or frozen depending on the member Confirm the chelator content of process water before scale-up

Two handling details cause most of the avoidable losses. Powders should be brought to room temperature before the vial is opened, because condensation on cold lyophilised cake is the usual reason a fresh lot underperforms on first use, and liquids should be aliquoted rather than repeatedly warmed and returned to the cold room. Neither is glamorous advice, and both are cheaper than a replacement order.

Tailored Esterases and Scale-up

— 07 — from a substrate you cannot name to a specified bulk item

Most enquiries start with a material rather than an enzyme name, which is the sensible order. Send the substrate, the acetyl content if you have it, and what you need the polymer to do afterwards, and the screening comes first; the catalogue is where the conversation ends, not where it begins.

  • Screening: our isolate collection is screened against the substrate you send, not against a reference polymer, which is how the unexpected matches turn up - a marine esterase that outperforms the textbook choice on an unusual acetyl pattern is a regular outcome.
  • Construct design: gene synthesis, host selection and construct building for expression in Escherichia coli, in Bacillus hosts, or in Komagataella where secretion into the broth simplifies recovery; the affinity tag is retained or cleaved according to what the protein has to do afterwards.
  • Stabilisation: formulation and protein engineering for the temperature and pH your line actually runs at, supported by half-life data rather than an optimum point.
  • Cocktails: esterases rarely work alone. We assemble and test combinations with hydrolases on real feedstock, then hold the blend as one specified item so repeat orders behave the same way.
  • Pilot and bulk supply: fermentation development and downstream capture, concentration and buffer exchange, ending in bulk powder, stabilised liquid or a filled presentation of your choosing.
  • Custom presentation: vial format, closure type, insert wording and paperwork matched to your own brand, so pallets can move straight into your warehouse without being repacked first.

Each project closes with the paperwork a process engineer needs: host and expression summary, purification route, activity assay protocol with both the chromogenic and the natural-substrate figure, electrophoretic purity evidence, contaminant screening, and the thermal data behind the stated storage conditions. Tell us the polymer, the substituent you need removed, and the quantity per campaign; the options worth testing are usually fewer than you expect.
